摘要
目的:探讨工作室内温度和使用时间及酶标抗体使用量对免疫印迹技术检测ENA多肽抗体谱的检测结果的影响。方法:试剂盒随着使用时间的延长对检测结果的影响,为了保证检测效果所使用酶标抗体的最适剂量。结果:平均工作室内温度有30℃,试剂失效最快,为5 0 d~6 0 d。平均工作室内温度有18℃失效较慢,为80 d~12 0 d;为了保证检测的效果达到最佳,新购买的试剂盒使用酶标抗体的剂量是15 μl。结论:随着使用开封试剂时间推延和随着室温的升高,有效期缩短;在使用试剂盒的过程中,使用酶标抗体的剂量要根据使用的时间和温度变化及试剂是否失效适当进行调节。
Objective To observe the effects of ind oo r temperature, time and the amount of enzymic labelling antibody in determinatio n ENA with immunblottingtechnique.Methods The inactivation of ENA kits was observed in different temperatures and duration. To assure the results , optimal dose of enzymic labelling antibody should be adjusted.Results At average indoor temperature 30℃,the reagent was inacted after 50~60 d a ys. At average indoor temperature 18℃, the reagent was inacted after 80~120 da y s. The optimal dose of enzymic labelling antibody should be adjusted as 15 μl . Conclusions With the time by and temperature raising,the duration of inactivation for ENA kits was shortened and the optimal dose of enzymic labellin g antibody should be adjusted.
