大白兔颈动脉移植后血浆一氧化氮浓度的变化 被引量:1

Changes of plasma nitric oxide level in rabbits of carotid transplantation model

摘要 目的: 探讨大白兔颈动脉移植后血浆一氧化氮(NO)浓度的变化及其意义。方法: 建立大白兔颈动脉移植模型,根据移植前供体血管不同处理方法将30只大白兔随机分为A(自体血管移植组)、B(无预处理异体血管移植组)、C(青霉素和链霉素处理后常温保存异体血管移植组)、D(青霉素和链霉素处理后液氮冷冻保存异体血管移植组)4组。观察移植后血管形态变化。测定并比较移植后1, 2, 3周血浆NO浓度。结果:B组血管移植后宿主血浆NO平均含量显著高于A组和D组(P<0.05);C组血浆NO含量和D组与A组无显著差异(P>0.05)。在血管形态上,A组结构正常,仅有炎性细胞浸润。其余各组术后1周可见内膜开始增生, 2周内膜明显增厚, 3周内膜增厚最显著。其中B组改变最严重。结论: 青霉素和链霉素可降低供体血管的抗原性及宿主诱导型一氧化氮合酶活性;液氮冷冻保存可能具有协同效应。 Objective: To investigate and assess the changes and significance of plasma nitric oxide (NO) in common carotid transplantation model of rabbits. Methods:We established an animal model of common carotid transplantation with rabbits in vivo. Thirty healthy homozygotic Japanese rabbits were randomizedly assigned into 4 groups (Group A, autografts; Group B, allografts without precondition; Group C, allografts precubated with penicillin/streptomycin and preserved under room temperature; Group D, preconditioned with penicillin/streptomycin and cryopreserved in liquid nitrogen). We observed the histomorphological changes of transplanted vessels and determined the concentration of plasma NO 1, 2 and 3 weeks after the transplantation. Results: The plasma NO concentration of rabbits in Group B was significantl higher than that in Group A and D (P<0.05). However, there was no significant difference between Group A and Group C, D (P>0.05). The vascular structure in Group A was normal, only with infiltration of inflammatory cells. In the other groups, the intima began to proliferate 1 week after the transplantation. Two weeks later, distinct thickening was observed, and the thickening reached the most in 3 weeks. The changes were found the most significant in Group B. Conclusion:Antigenicity of allografts and activities of induced nitric oxide synthase might be down-regulated by precondition with both or either of penicillin and streptomycin. Cryopreservation might have synergetic effect.
