摘要
目的: 通过观察大鼠原位肝移植全肝缺血期血液动力学、心电图、胃肠淤血、游离血红蛋白变化,探讨自制转流泵在大鼠原位肝移植模型中的应用。方法:根据滚压式转流泵的工作原理,将卡带单放机改制成小型转流泵应用于大鼠全肝血流阻断门静脉转流期。20只大鼠行全肝阻断后,行脾静脉股静脉转流。比较阻断前、阻断后、单纯分流或应用转流泵转流5, 10, 15min后血液动力学指标,同时监测心电图变化及测定应用转流泵转流后游离血红蛋白含量。结果:与单纯分流比较应用转流泵后大鼠血液动力学稳定,门脉系统回流通畅;心律失常发生率明显减少;转流后游离血红蛋白含量在临床可接受的正常范围内。结论:自制转流泵可以应用于大鼠原位肝移植模型的建立。
Objective: To investigate the changes of hemodynamics, electrocardiogram(ECG), stasis of stomach and intestines, and the content of dissociated hemoglobin during the hepatic ischemia in order to determine whether a self-made rotary pump can be used in the model of hepatic original transplantation or not. Methods: According to principle of field roller rotary pump, a tape recorder was made into a small rotary pump, and was used in the period of portal blood bypass while the hepatic inflow was obstructed. Blood of livers of 24 male Wistar rats were blocked. A bypass was used between spleen vein and thigh vein. The changes of ECG, mean arterial pressure(MAP), and heart rate(HR) were recorded before obstructed, when obstructed, with simple shunt, and with bypass for 5, 10, 15 minutes after using rotary pump. Results: The hemodynamic of using rotary pump were more stable than that of using the simple shunt. Less stasis of portal vein and less arrhythmia were also observed after portasystemic bypass. The contents of dissociated hemoglobin were also at the normal range after bypass. Conclusion: Self-made bypass pump can be used in the establishment model of hepatic original transplantation of rats.
