日本鬼鲉毒素对大鼠免疫功能的影响

摘要 实验选用大鼠腹腔巨噬细胞(PM )为细胞模型,观察日本鬼鲉(Inimicus japonicus)毒素在体外对免疫细胞功能和结构的影响,扫描电镜观察毒素对巨噬细胞形态的影响,并检测注射毒素局部组织过氧化物的含量,应用维生素C(Vc)探讨其对毒鱼蛰伤有无治疗或阻断作用。结果表明,日本鬼鲉毒素可抑制大鼠腹腔巨噬细胞吞噬中性红功能,MTT活性明显降低,腹腔巨噬细胞P物质(Substance P, SP)表达明显降低;通过电镜观察到毒素损伤巨噬细胞结构,Vc治疗组有部分改善;毒素注射局部皮肤组织中MDA 含量明显升高,而抗氧化GSH -Px、SOD 活性和总抗氧化能力(T -AOC)显著降低,Vc治疗组MDA含量显著降低,抗氧化酶活性和总抗氧化能力提高。结果提示,被日本鬼鲉蛰伤后局部组织容易感染和难愈合的原因可能与免疫细胞活力下降密切相关。抗氧化剂Vc可以减轻毒素的作用,毒鲉科棘毒鱼蜇伤的机制可能部分与氧化损伤机制有关。 We observed the effects of the venoms of Inimicus japonicus on the phagocytosis and activity of the macrophagocytes of the abdominal cavities in vitro. The structure of the macrophagocytes was observed by scanning electron microcope. The content of MDA and the activities of GSH-Px, SOD, and T-AOC in the skin of injection were detected. We further investigated the preventive function of VC on the skin damage. The phagocytic function of macrophagocytes decreased. The morphological changes of the macrophagocytes were observed by scanning electron microcope. The venoms could increase MDA content and decrease the activities of GSH-Px, SOD, and T-AOC in the skin injected with venoms. VitC (inhibitor of reactive oxygen species) had a preventive function to reduce the damage in some degree. These indicated that the damage was mainly caused by the decreasing activity of the immunity and reactive oxygen species.
