摘要
目的 探讨鼠神经细胞和巨噬细胞在体外的联合培养条件 ,为研究神经系统疾病的炎症发病机制奠定实验基础。方法 取大鼠小脑的神经细胞和小鼠腹腔的巨噬细胞进行联合培养 ,观察联合培养细胞的生长特性 ,采用免疫组化法检测神经细胞的微管相关蛋白 - 2 (microtubule -associatedprotein 2 ,MAP - 2 )。 结果 大鼠小脑的神经细胞和小鼠腹腔的巨噬细胞联合培养后不影响各自的生长状况和形态结构 ,神经细胞的MAP - 2免疫组化显示神经细胞的形态无明显变化。结论 鼠神经细胞和巨噬细胞在体外培养条件下可以共生长 ,这一联合培养的方法可以作为神经系统疾病研究的一种新的实验方法。
Objective To explore the culturing condition of the neuron of rats and macrophage of mice in vitro and search a new mean of experiment for investigating immune-mediated neural diseases. Methods The cerebellar granule neurons of rats and the peritoneal cavity macrophage of mice were co-cultured to observe their living speciability . Microtubule-associated protein 2 (MAP-2) of the neuron was detected by immunocytochemical technique.Results The growth and MAP-2 expression of co-cultured neuron and macrophage were not affected.Conclusions The neuron and macrophage could co-grow in vitro ,which may provide a key insight for immune-mediated neural diseases.
