摘要
目的通过观察大鼠局灶脑缺血预处理模型对钙依赖中性蛋白酶(calpain)活性变化的影响,初步探讨calpain与脑缺血预处理神经保护作用的关系。方法55只Wistar大鼠随机分为4组,假手术对照组、预处理对照组、预处理缺血组、脑缺血组,比较各组神经功能评分、脑梗死体积、calpain活性变化及组织病理变化。结果预处理缺血组神经功能评分、梗死体积明显低于脑缺血组(P<0.01)。预处理缺血组calpain活性低于脑缺血组(P<0.05)。组织病理可见预处理缺血组有部分梗死性病变,神经元数量明显减少,但组织结构轮廓尚存。假手术对照组及预处理对照组无神经功能缺损及梗死灶形成。结论脑缺血预处理可产生缺血耐受现象,减小梗死体积,减轻缺血性神经损伤,抑制calpain活性。calpain活性减弱可能在脑缺血耐受中发挥一定作用。
Objective To investigate the effect of focal cerebral ischemic preconditioning on the calpain activity and to explore its role in ischemic tolerance in rats. Methods Fifty-five Wistar rats were randomly divided into four groups:sham-operation control group, cerebral ischemic preconditioning control group, cerebral ischemic preconditioning group, and cerebral ischemia group. Neurologic deficit scores, infarct volume, HE staining and changes of calpain activities were evaluated in each group. Results Compared with cerebral ischemia group, there was lower neurologic deficit scores, a smaller volume(P<0.01), less severe neuronal injury(HE staining) and lower calpain activities(P<0.05) in cerebral ischemic preconditioning group. No neurologic impairment and infarct were identified in the sham-operation control group and cerebral ischemic preconditioning control group. Conclusions Ischemic preconditioning is strong enough to induce ischemic tolerance and to attenuate activity of calpain.
