摘要
目的 研究脑出血(ICH)后血肿周围区细胞凋亡的时间规律及胞磷胆碱钠治疗对血肿大小、细胞凋亡的影响。方法 自体非抗凝动脉血注入大鼠右侧尾壳核制作脑内出血模型;腹腔注射胞磷胆碱钠或生理盐水;按多田公式估算血肿体积;流式细胞仪检测血肿周围区细胞凋亡率。结果 大鼠脑内出血后6 h 血肿周围区出现细胞凋亡,72 h达高峰,4周时有少量凋亡细胞存在;对应的相同时间点给药组与对照组大鼠脑内血肿体积比较无显著差异(P>0.05);除6 h外余各时间点均显示给药组细胞凋亡率低于对照组,尤以72 h组为甚(P<0.01)。结论 脑内出血后血肿周围脑组织发生细胞凋亡且有时间规律性;胞磷胆碱钠早期治疗实验性大鼠脑内出血不引起血肿扩大,且可减少血肿周围区细胞凋亡。
Objective To study cell apoptosis and its time course in the area around the hematoma after intracerebral hemorrhage (ICH), as well as the influence of citicoline sodium on it.Methods ICH was induced in adults rats by stereotaxical injection of autologous caudate artery blood into right caudate nucleus. Citicoline sodium 500 mg/kg or saline was administered intraperitoneally. The volume of hematoma was determined by Tada formula. The time course of cell apoptosis and the effect of citicoline sodium on it were determined by the dynamic change of apoptosis rate measured by flow cytometry (FCM).Results In the area around intracerebral hematoma in rats, cell apoptosis occurred at 6h after the onset of ICH, peaked at 72h, then descended and lasted for 4 weeks. There was no difference in the underlying hematoma volumes when animals were treated with citicoline compared with placebo treated animals at the same time. In terms of the rate of cell apoptosis around hematoma, there was difference at every phase between the citicoline treated group and the placebo treated group, except at 6h. Especially at 72h, were the group treated with saline markedly more than that of the group treated with citicoline, showing significant difference ( P < 0.01 ).Conclusions ICH can trigger cell apoptosis in the area around intracerebral hematoma in rats, and there is regularity at time when apoptosis occurs Early treatment with citicoline sodium in experimental intracerebral hemorrhage can reduce the cell apoptosis in the area aroune the hematoma and can not increase the volume of the intracerebral hematoma.
