Survivin、p53和bcl-2在脑星形细胞肿瘤中的表达及相关性研究

摘要 背景与目的:近年来发现胶质瘤生长、分化有着其特定的分子机制,胶质瘤恶性程度与特定基因的突变、丢失或扩增表达有密切的关系。本研究探讨凋亡抑制基因Survivin、抑癌基因p53和癌基因bcl-2在人脑星形细胞肿瘤中的表达、相互关系及其意义。方法:按制定的人组标准选择45例脑星形细胞肿瘤标本,应用免疫组化技术检测Survivin、p53、bcl-2在不同级别脑星形细胞瘤中的表达。结果:45例脑星形细胞肿瘤标本中33例表达Survivin蛋白,阳性率为73.3%,并随星形细胞肿瘤病理分级的增加而增高(P<0.05);胶质母细胞瘤的p53阳性水平要高于星形细胞瘤:bcl-2阳性水平星形细胞瘤显著高于胶质母细胞瘤;Survivin表达与 p53表达密切相关,与bcl-2表达无相关性。结论:Survivin高表达与星形细胞肿瘤的恶性进展有关;Survivin的表达与脑星形细胞瘤中p53异常表达密切相关。 BACKGROUND & OBJECTIVE: Tumorigenesis and tumor progression have been suggested to be related to abnormal gene expression. This study was designed to examine the expression of survivin, p53 and bcl-2 gene in human brain astrocytic tumors and the relationship among these three proteins. METHODS: Fortyfive specimens of astrocytic tumors classified according to 1993 WHO Grade system were selected and used in this study. The samples were immunohistochemistry stained using streptavidinbiotin peroxidase (SP) technique to determine the expression of survivin, p53, bcl-2 protein. RESULTS: Of the 45 investigated tumors, according to 1993 WHO standard, 11 were astrocytomas (AS, grade II ), 16 were anaplastic 3astrocytomas (AA, grade Ⅲ) and 18 were glioblastomas(GBM, grade IV). Survivin protein expression was observed in 33 of 45 (73.3%) cases, and the positive rate was increased associated with tumor grade getting higher. The P53 expression in GBM was higher than that in AS, whereas most astrocytic tumors were positive for bcl-2 protein expression. The survivin expression was significant associated with p53 expression (P<0.05), but not with bcl-2 expression. CONCLUSION: The strong relationship between malignant grade and survivin expression indicates that expression of survivin may relate to tumor malignant progression of astrocytic tumors.
