Influence of Fluvastatin on bone morphogenetic protein-7 in the process of fracture healing of rats with osteoporosis

摘要 目的探讨氟伐他汀对骨质疏松大鼠骨折愈合过程中骨形态发生蛋白7(BMP7)的影响。方法6个月龄雌性SD大鼠96只,随机分为去势组和非去势组,每组48只,分别行双侧卵巢切除术和假性手术;3个月后,去势组于股骨中段制造骨折模型,再随机分为给药组和对照组,每组24只,给药组手术次日给予10mg/(kg·d)氟伐他汀灌胃,疗程为2w,对照组给予安慰剂。利用免疫组化及原位杂交检测骨折愈合过程中给药组和对照组骨折后3、7、14、21、28和42d骨痂中BMP7、BMP7mRNA的表达,并用真彩色图像分析仪作图像分析。结果给药组和对照组在骨折愈合过程中BMP7及BMP7mRNA基因表达的细胞定位无差别,且表达模式基本相似。术后14d,阳性细胞的阳性程度给药组比对照组强(P<0.05)。结论氟伐他汀对骨质疏松大鼠骨折愈合过程中BMP7有一定促进作用。 Objective To explore the influence of Fluvastatin on bone morphogenetic protein-7 (BMP-7) in the process of fracture healing of rats with osteoporosis. Methods 96 female SD rats aged six-month old were randomly divided into two groups: bilateral ovariectomized group (48 cases) and pseudo-operated group (48 cases). Three months later, fracture model was made on the mid-femoral in bilateral ovariectomized group, which were then randomly divided into administration group (24 cases, given 10 mg·kg^-1 ·d^-1 Fluvastatin, 2 weeks) and control group (24 cases, given placebo). The expression of BMP-7 and BMP-7 mRNA in the bone callus of both groups was examined by immunohistochemical method and hybridization in situ on the 3rd, 7th, 14th, 21st, 28th and 42nd days in healing process. Meanwhile image analysis was conducted with the color image analytical tool. Results There was no difference between two groups in the cellular localization of the expression of BMP-7 and BMP-7 mRNA gene with basically similar expression model. But after 14 days, the positive level of positive cell of the administration group was stronger than that of control group (P<0.05). Conclusions Fluvastatin should have certain promotive effect on bone morphogenetic protein-7 (BMP-7) in the process of fracture healing of rats with osteoporosis.
