摘要
目的检测相关细胞因子血管生成素1(Ang1)、Ang2在大鼠局灶性脑缺血/再灌注后表达的动态变化。方法采用免疫组化及原位杂交技术。结果在正常脑组织及缺血/再灌注后均发现Ang1蛋白及Ang1mRNA的中等程度的广泛表达,在再灌注48h时达到高峰,此后虽有所下降,但仍为持续的阳性表达。Ang2蛋白及Ang2mRNA在对照组及假手术组中均呈阴性表达,在MCAO后6h于梗死灶内的一些单个细胞中发现,在12~24h后达到高峰,这种表达一直持续到72h。结论Ang1、Ang2在脑损伤的过程中起着重要的作用。
Objective To measure dynamic changes in mRNA expression of the related factors, Angiogenesis (Ang)-1, Ang-2 after focal cerebral ischemia-reperfusion of rats. Methods Immunohistochemistry and in situ hybridization technique were adopted. Results Moderate expressions of both Ang-1 protein and mRNA were commonly found in normal cerebral tissues and those after ischemia-reperfusion, which reached peak at 48 hours after reperfusion, then declined little while still remained positive. Both Ang-2 protein and mRNA were negative expressions in control group and pseudo-operation group, but were found in some isolated cells of infarction focus at 6 hours after MCAO, and reach peak at 12~24 hours after MCAO and lasted for 72 hours. Conclusions Ang-1 and-2 could play the important role in the course of cerebral injury.
