摘要
目的:为移植免疫及药物免疫机理研究建立小鼠同种异基因皮肤移植动物模型。方法:受体BALB/c分为A~E共5组,即对照组,5 0、7 5、10、2 0mg/kg环孢菌素A(CsA)剂量组,通过背-背法进行C5 7BL/ 6的全厚皮皮片移植,以探求通过较小剂量CsA及优化实验条件和护理方式建立供免疫机理研究的动物试验平台。结果:5 0、7 5、10mg/kg组相对于对照组皮片排斥时间无明显差别,而2 0mg/kg组植皮能较长时间存活。结论:2 0mg/kg的CsA剂量可以经济。
Aim: To establish a model of allogenetic skin transplantation animal in mice so as to carry out further studies on the immune mechanism of transplantation and immune-effect of traditional Chinese medicine. Methods: Recipient BALB/c are divided into five groups (A, B, C, D and E), full-thickness skins are transplanted through back to back from donor (C57BL/6.) Corresponding doses of cyclosporin A (CsA) (A:control, B:5.0 mg/kg, C:7.5 (mg/kg), D:10 mg/kg, E:20 mg/kg) are administrated through intraperitoneal infusion, different experimental conditions are studied to optimize our proceeds of establishing of animal model to provide a animal trial stage for the studies of immuno-mechanism of drugs and transplantation. (Results:) The times of graft-rejection in group 5.0, 7.5, 10 mg/kg have no obvious difference with the control group A while the grafts of 20 mg/kg CsA group survived for a long time. (Conclusion:)Observable model could be economically established with dose of 20 mg/kg CsA.
