摘要
目的: 通过研究双歧杆菌表面分子脂磷壁酸(LTA)对衰老小鼠肝脏形态、p16蛋白、c Fos蛋白表达及骨髓细胞微核率的影响,探讨双歧杆菌抗衰老的分子机制.方法:雄性昆明系小鼠,随机分组,在建立D 半乳糖诱导衰老小鼠模型的同时,每日ip双歧杆菌LTA. 40d后,采用免疫组化SABC法、Gimesa染色和其他它方法检测小鼠体质量、肝脏指数、形态、p16蛋白、c Fos蛋白及骨髓红细胞微核率的变化.结果: 与青年对照组小鼠相比,衰老模型组小鼠体质量、肝脏c Fos蛋白表达明显降低(P<0 05),p16蛋白表达及骨髓红细胞微核率显著增高(P<0 05);给予双歧杆菌LTA,衰老模型小鼠体质量与肝脏c Fos蛋白水平明显增高(P<0 05),而p16蛋白水平及骨髓红细胞微核率显著降低(P<0 05),并且肝脏组织形态结构也得到了明显改善.结论: LTA可延缓肝脏与骨髓衰老,抑制染色体畸变,上调c fos,下调p16表达.
AIM: To study the effect of lipoteichoic acid (LTA) of bifidobacterium on liver and bone marrow in aging mice and to explore its mechanism.METHODS: Aging mice were induced by sc D- galactose and LTA was simultaneously given ip to them daily,and for 40 days.The body mass,liver index,liver morphology,the expression of p16,c-fos protein and micronucleus frequency in the bone marrow were tested by Gimesa staining,SABC technique of immunohistochemistry and other methods.RESULTS: Compared with those in the young control group,the body mass and the expression of c-fos protein in liver noticeably reduced in aging model mice(P<0.05)while the expression of p16 protein in liver and micronucleus frequency in the bone marrow remarkably increased (P<0.05).After administrating LTA to aging model mice,all the above mentioned changes were reversed(P<0.05).CONCLUSION: LTA of bifidobacterium has an effect of deferring senescence of liver and bone marrow in aging model mice.
