摘要
目的: 探讨Fas基因表达水平与胃癌细胞增殖活性及细胞凋亡程度的关系.方法: 胃癌组织53例,用原位杂交及免疫组化染色法分别检测FasmRNA,Fas蛋白和增殖细胞核抗原(PCNA)的表达,并采用凋亡细胞原位检测方法对组织切片中的凋亡细胞进行观察和比较.结果: 胃癌组织表达FasmRNA28例( 52 8% ),表达Fas蛋白24例( 45 3% ),两种方法检测结果一致性非常显著(P<0 01).胃癌53例增殖期细胞标记物PCNA表达及凋亡细胞的阳性率均为100%,细胞凋亡和细胞增殖指数呈显著性负相关(r= -0 982,P<0 01).随胃癌细胞Fas蛋白表达水平升高,PCNA阳性细胞指数相应减少,肿瘤凋亡细胞指数则相应增加.Fas蛋白组细胞凋亡指数显著高于- /+组(P<0 01 ),Fas蛋白组PCNA指数显著高于 / 组(P<0 01 ).结论: 胃癌细胞Fas基因表达可引起细胞凋亡增加与增殖减少.
AIM: To investigate the relationship between Fas gene expression level in human gastric carcinomas and the frequency of cell proliferation activity and the apoptosis of the tumor cells.METHODS: In situ hybridization and immunohistochemistry methods were used to study the frequencies of expression of Fas gene and proliferating cell antigen (PCNA) in 53 gastric carcinomas.An in situ apoptotic cell detection method (TUNEL method) was also adopted to detect the apoptotic cells.The number of apoptotic cells and PCNA cells was compared with the Fas protein expression in each case.RESULTS: Of the 53 gastric carcinomas,28(52.8%) and 24(45.3%) expressed Fas mRNA and Fas protein respectively.There was no significant difference in the positivity rates obtained by these 2 methods.The apoptotic index and PCNA index of gastric carcinomas were correlated negatively (r=-0.982,P<0.01).With the increase in the expression of Fas protein,the cell proliferating activity decreased and apoptosis increased in the tumor cells.There was significant difference in apoptosis indices between Fas protein group and -/+ group (P<0.01).The PCNA index in Fas protein group was significantly higher than that in and groups(P<0.01).CONCLUSION: Fas gene can promote apoptosis and inhibit cell proliferation in the development and malignant progression of gastric carcinomas.
