摘要
目的: 探讨三氧化二砷(As2O3 )对裸鼠移植人乳腺浸润性导管癌组织中NF κBp65,p53,c Myc和hTERT蛋白表达的影响.方法: 复制人乳腺浸润性导管癌裸鼠移植瘤模型22例,随机分为4组.用免疫组化法检测上述组织中各蛋白的表达.结果: NF κBp65,p53,c Myc和hTERT蛋白在人乳腺浸润性导管癌裸鼠移植瘤阴性对照组中的阳性表达均高,且有一致性;实验组1、实验组2的p65阳性表达随着As2O3 剂量的增加而下降(P<0 01),与阳性和阴性对照组的表达比较呈明显下降趋势(P<0 01).p53,c Myc和hTERT蛋白在四组中的表达无差异性(P>0 05).结论: As2O3 通过抑制核因子NF κBp65的活性抑制人乳腺浸润性导管癌细胞的生长作用,而对p53,c Myc和hTERT蛋白无影响.
AIM: To investigate the effect of As 2O 3 on the expressions of NF-κB p65,p53,c-myc and hTERT protein in heterologous graft model for human breast infiltrating duct carcinoma in nude mice.METHODS: A model of nude mice bearing human breast infiltrating duct carcinoma was set up and divided randomly into four groups.The expressions of NF-κB p65,p53,c-myc and hTERT protein were examined in the tissues by immunohistochemistry.RESULTS: The positive expression rates of NF-κB p65,p53,c-myc & hTERT protein were high in the negative group of nude mice model.The positive expression of NF-κB p65 in experimental group 1 and 2 was gradually descending with the increase dosage of As 2O 3 (P<0.01) and showed a remarkable descending tendency compared with that in the negative and male groups (P<0.01).But the expressions of p53, c-myc and hTERT protein were not different among the four groups (P>0.05).CONCLUSION: As 2O 3 inhibits the high expression of NF-κB p65 protein in heterologous graft model for human breast infiltrating duct carcinoma in nude mice in a dose-dependent manner and As 2O 3 does not affect the expression of p53,c-myc and hTERT protein.No relationship is found between NF-κB p65,p53,c-myc and hTERT.
