摘要
以丹参(SalviamiltiorrhizaBunge)幼嫩叶片为外植体,在MS+2,4-D0.5mg/L+6BA1.5mg/L培养基上诱导形成愈伤组织,愈伤组织置于不同条件下培养。探讨丹参细胞培养过程中蛋白质、酶活性的变化与细胞生长周期之间的关系。结果显示黑暗和光照下,培养细胞的生长周期为27天;可溶性蛋白含量和超氧化物歧化酶(SOD)活性均出现两个明显峰值,而过氧化物酶(POD)活性变化较复杂。丹参细胞生长呈"S"形周期性变化,其可溶性蛋白、酶活性也呈相应的周期性变化。
The relationship between the changes of soluble protein content and activities of enzymes were studied.Based on young leaves of Salvia miltiorrhiza Bunge,callus was induced on the solid medium of MS+2,4D(0.5mg/L)+6BA(1.5mg/L).Two groups of callus were cultured in the dark and light respectively.The influeces of light on the cell growth and physiological and biochemical characteristics were tested.The growth circles were about 27 days.Soluble protein content and superoxide dismutase(SOD) showed two clear peaks while activities of peroxides(POD) had complexing changes. The result showed that the cell growth curves were like'S'.The soluble protein content and activities of enzymes relevantly had periodic changes.
