反义寡核苷酸对骨肉瘤细胞组织蛋白酶L表达的影响

摘要 目的 观察合成人组织蛋白酶L(CATL)基因的反义脱氧寡核苷酸转染后对人骨肉瘤MG 63 细胞系中CATL表达的影响。方法 人工合成正义、反义及无意义CATL基因片段,转染人骨肉瘤细胞MG 63,免疫细胞染色观察CATL的表达,RT PCR和Western blot检测CATL的mRNA和蛋白质表达水平的变化。结果 CATL反义寡核苷酸对MG 63 细胞的CATL的表达有明显的抑制作用,与正义链组、无意义链组和空白对照组相比有显著差异(P<0.05)。结论 反义寡核苷酸的转染可阻遏骨肉瘤细胞中CATL的表达。 Objective To study the effect of CATL phosporothioated antisense oligodeoxynucleotide on CATL expression in human osteosarcoma cell line MG-63.Methods The antisense,sense and nonsense oligodeoxynucleotides for cathepsin L gene were synthesized and delivered into the human osteosarcoma cell line MG-63.The effects of oligodeoxynucleotides were examined with immunohistochemistry,RT-PCR and Western blot.Results The expression of cathepsin L was remarkably decreased after antisense oligodeoxynucleotide treated cells compared with the control cells.Conclusion The cathepsin L gene antisense oligodeoxynucleotide can significantly inhibit the expression of cathepsin L in MG-63 cell.
