摘要
目的探讨肾安胶囊对肾大部切除大鼠肾脏功能的影响。方法将大鼠随机分为假手术组和手术组,术后3天再将手术组大鼠随机分为肾安胶囊组和Nx组。8周后测尿蛋白和血肌酐;取肾组织做病理检查;用免疫组织化学检查肌成纤维细胞标志抗原(α-平滑肌肌动蛋白α-SMA)。结果Nx组大鼠肾脏出现肾小球硬化和肾间质纤维化;和Sham组相比α-SMA表达明显升高;肾安胶囊组肾脏纤维化明显减轻(P<0.05),α-SMA表达比Nx组明显下降。结论肾安胶囊可抑制肌成纤维细胞增殖和浸润,减轻肾脏纤维化从而保护肾功能。
Objective To investigate the effect of Shen'an capsule on renal fibrosis in 5/6 renal ablation in rats. Methods The rats were randomly divided into Sham group and nephrectomy group. In the 3rd day after surgery nephrectomy rats were randomly subdivided in two groups of shenan and Nx and Sham rats were received equal volume water by daily gastric lavage. All rats were allowed free access to food and water. In 8 weeks after the treatment with Shen'an capsule, the following studies were performed in every group. All of the items such as body weight, the amount of 24 hours urine protein, serum creatinine(Scr) were measured. The renal tissue of three group rats was observed with light microscope after periodic acid-Schiff (PAS) and Masson staining. Immunohistochemical staining was used to estimate the expression of α-SMA in renal cortex and the results were analyzed semi-quantitatively with the pathological image analysis system. Results Expression of α-SMA was increasingly strong in the glomeruli, tubules and interstitium of remnant kidneys in Nx rats during eight weeks. Their expression was reduced significantly with Shen'an capsule treatment (P<0.05). The histologic observations are shown severe mesangial expansion, extensive capillary loops ollusion, tubular atrophy and interstitial fibrosis in eight weeks in Nx group, but the abnormal changes were alleviated in Shen'an group. Conclusion Treatment with Shen'an capsule markedly inhibits proliferation of myofibroblast. It has protective effect on kidney by preventing and delaying the renal fibrosis.
