摘要  目的:观察桂附地黄丸对老年狗骨髓基质干细胞增殖能力的影响。方法:取4只老年杂种狗,给予桂附地黄丸口服,用药前后抽取骨髓进行细胞分离,并以细胞密度为1×105/mL、1×106/mL、1×107/mL接种,观察细胞生长情况。并对获得的细胞进行脂肪诱导、骨诱导分化试验。结果:4只狗服药前的骨髓接种后,细胞不易生长、传代;服药后骨髓细胞较易生长及增殖。生长的细胞进行的成脂肪和成骨诱导分化试验为阳性结果。结论:桂附地黄丸对老年狗骨骨髓基质干细胞的增殖具有一定促进作用。 Aim To observe the effect of a Chinese traditional medicine - Gui Fu Di Huang bolus on the cellular proliferation of the senile dog bone marrow mesenchymal stem cells (MSCs). Methods Four senile hybrid dogs were involved in this experiment and the boluses were given orally 1 pill a day for 20 days to the dog. Before and after the boluses applied, bone marrow mesenchymal mononuclear cells were extracted, isolated and cultured at the density of 1×10\+5/ml, 1×10\+6/ml, 1×10\+7/ml in 24 well plates. Osteogenic differentiation test and adipogenic differentation test were performed to detect the multilineage differentiation ability of the obtained cells. Results: After the treatment, the cells could grow and reexpand whereas the cells obtained before the boluses applied were difficult to grow. The cells of post-treatment cultured for multilineage differentation is positive to oil red-O staining, and positive to alizarin red staining, Von Kossa staining and Alkaline phosphatase staining after osteogenic differentiation. Conclusion Gui Fu Di Huang bolus can promote the proliferation of the senile dog bone marrow mesenchymal stem cells(MSCs).
