中国枪乌贼墨汁提取物的抗肿瘤作用

Antitumor effects of extract from the ink of Loligo chinensis

目的:研究中国枪乌贼墨汁提取物(以下简称墨汁提取物)的抗肿瘤作用。方法:采用超声波处理、低温离心、过滤等方法制备墨汁提取物;台盼蓝染色法检测墨汁提取物对体外培养的人髓性白血病细胞(HL-60)、人鼻咽癌细胞(CNE-2Z)和人子宫颈癌细胞(HeLa)生长的抑制作用;采用体内肿瘤实验技术研究墨汁提取物对小鼠移植性肉瘤180(S180)和肝癌(H22)生长的影响;用常规方法进行墨汁提取物的急性毒性试验。结果:提取物对HL-60,CNE-2Z和HeLa细胞都有明显的抑制效果,3种细胞中以HL-60细胞最敏感;墨汁提取物腹腔注射1.2g·kg-1和灌胃2.4g·kg-1共15d对S180的抑制率分别为70.8%和69.2%,对H22的抑制率分别为71.0%和58.0%;LD50测不出,最大耐受量>48g·kg-1。结论:中国枪乌贼墨汁提取物在体内外都有显著的抗肿瘤作用,且无明显的急性毒性。

Objective To study the antitumor effect of extract from the ink of Loligo chinensis. Methods The extract from the ink of Loligo chineusis was isolated by ultrasound technique and low temperature centrifugation. Growth inhibition of tumor cells was routinely estimated by trypan blue staining in vitro,and in vivo its antitumor effects were investigated by testing the tumor weight of tumor-bearing mice (S 180 and H 22). Results The extract from the ink of Loligo Chinensis display obvious growth inhibitory effect on HL-60,CNE-2Z and HeLa cells. When administerd ip or ig 24g·kg for 15 d,the extract caused 70.8% and 69.2% inhibition rates on S 180, 70.5% and 58.1% inhibition rates on H 22, respectively. In addition, the maximum tolerance dose of the extract was above 48 g· kg~ -1(the amount corresponds to that of the natural ink of Loligo chineusis), and the LD 50 could not be obtained.Conclusion The extract from the ink of Loligo chinensis showed potent antitumor activity and low toxicity.
