平肝汤抗大鼠肝纤维化的实验研究

Experimental Study on the Effects of Pinggan Tang on Liver Fibrosis

目的观察平肝汤对CCl4诱导的大鼠肝纤维化的防治作用。方法64只雄性大鼠随机分为空白组、模型组、秋水仙碱组及平肝汤组,每组16只。除空白组外,所有大鼠皮下注射40%CCl4花生油溶液,每隔3d1次,共12周。空白组予以等量的花生油皮下注射。从造模第4周起,各组给予相应的药物或生理盐水灌胃。实验结束后采集血清、肝脏组织标本,检测血清ALT(丙氨酸氨基转移酶)、AST(天门冬氨酸氨基转移酶)、Alb(白蛋白)、Glo(球蛋白)、PCⅢ(Ⅲ型前胶原)、Ⅳ型胶原(Ⅳ-Co)、LN(层粘连蛋白)、HA(透明质酸),观察肝组织学改变。结果平肝汤组、秋水仙碱组大鼠肝组织纤维化病变较模型组轻(P<0.05),血清ALT、AST、Glo、PCⅢ、Ⅳ-Co、LN、HA亦较模型组低(P<0.01),而Alb水平高于模型组,平肝汤组疗效显著优于秋水仙碱组(P<0.05)。结论平肝汤对CCl4所致大鼠肝纤维化有良好的防治作用,具有保护肝细胞、减轻肝细胞变性坏死、抑制肝脏纤维组织增生、减轻肝组织学病变的作用。

Objective To study the effects of Pinggan Tang on liver fibrosis. Methods Sixty four Wistar rats were randomly divided into four groups:control, model, colchicine, Pinggan Tang。All rats, excepting rats in control group, were given subcutaneous injection of 40% carbon tetrachloride (one time every three days). The rats in control group were given subcutaneous injection of peanut oil. Since the fourth week of the test, all rats were given medicine via gastrogavage respectively. At the end of 12th week, all rats were sacrificed and serum levels of hyaluroni acid (HA), laminin(LN), procollagen type-Ⅲ (PCⅢ), collagen type-Ⅳ(Ⅳ-Co), Alanine aminotransferase (ALT), Asparagine aminotransferase (AST), albumin(Alb), globulin (Glo) were measured. The histological changes of liver tissue were observed with light microscope. Results Compared with model group, the serum levels of HA, LN, PCⅢ, Ⅳ-Co, ALT, AST, Glo in colchicines and Pinggan Tang groups were lower, and the liver histological changes in rats of both colchicines and Pinggan Tang groups were less than control group. The damage in rats of Pinggan Tang group were least. Conclusion Pinggan Tang is very efficient in preventing and treating liver fibrosis.
