实验性热损伤所致的兔耳中央动脉血管平滑肌细胞增生

摘要 目的 探索建立热刺激动脉诱发血管平滑肌细胞增生的动物模型。 方法 兔耳中央动脉中间2cm处被热水(4 3±0 .5 )℃刺激,每天一次,每次3h ,共做3次。分别于造模后3d、1周、2周、4周时取材,石蜡切片。光学显微镜及图象分析仪进行形态学观察。 结果 中膜成分增生并突入血管腔内,造成管腔变狭。造模4周时管腔面积与造模3d时、造模7d时相比明显缩小(2 .5 8±0 .3 5 )mm2 /10 0比(3 .2 5±0 .5 9)mm2 /10 0、(3 .14±0 .2 4)mm2 /10 0 ,P <0 .0 5。造模4周时中膜厚度及平滑肌细胞层数与造模3d时、造模7d时相比明显增加(10 0 .5±15 .5 ) μm比(69.2±16.1) μm、(71.4±16.3 ) μm ,P <0 .0 5 ;(11.6±0 .8)层比(7.2±1.1)层、(6.5±1.7)层,P <0 .0 5。 结论 血管外热刺激可以诱发血管平滑肌细胞增生。该模型简便易行、重复性好。 Objective To build a new animal model of the vascular smooth muscle cells proliferation induced by thermal injury in the rabbit central ear artery. Methods The central artery of the rabbit ear was stimulated by warm water ( 43 ± 0.5 ℃ ) at the middle 2 cm for 3 hours daily for 3 days. Animals were killed at different time points, and tissue sections were examined under light microscope and digital image analysis system. Results The central artery of the rabbit ear responded to warm water, including the vascular smooth muscle cells proliferation, migration, and extending to the lumen of the artery. The lumen areas were significantly reduced 28 days after thermal injury compared with 3 or 14 days after thermal injury(2.58 ± 0.35 mm 2 / 100 vs 3.25 ± 0.59 mm 2 / 100, 3.14 ± 0.24 mm 2 / 100,P< 0.05 ). The thickness of the tunuca media and the number of the tunuca media smooth muscle cell both increased significantly 28 days after thermal injury compared with 3 or 14 days after thermal injury (100.5 ± 15.5 μm vs 69.2± 16.1 μm , 71.4 ± 16.3 μm,P < 0.05; 11.6 ± 0.8 layer vs 7.2 ± 1.1 layer, 6.5 ± 1.7 layer,P < 0.05, individually) respectively. Conclusion The vascular smooth muscle cells proliferation can be induced by thermal injury in the rabbit central ear artery. Owing to its high sensitivity, accuracy and convenience, the rabbit ear model is available for smooth muscle cell proliferation investigation.
