摘要
研究了基本培养基、碳源、营养添加物、pH值、固化剂等几种因素对日本结缕草‘Qingdao’和‘Zenith’两个品种成熟种子为外植体诱导愈伤组织的影响。结果表明:蔗糖作为碳源、pH值5.4~6.0、以植物凝胶或脱乙酰吉兰糖作为固化剂有利于日本结缕草愈伤的诱导与生长,NMB培养基、NB培养基分别有利于Zenith及Qingdao胚性愈伤的形成;而营养添加物的加入虽可提高愈伤诱导率,却导致胚性愈伤率的下降。同时,根据愈伤组织生长曲线得出,愈伤组织最适继代周期为24~33天。
Effects of several factors on callus induction and growth of zoysia japonica were studied.Mature seeds of two cultivar,'Qingdao'and'Zenith'were used to induce callus.The results showed that it was proper to use sucrose as carbon source,phytagel or gelrite as gelling agent,and pH value should be 5.4 to 6.0;NMB and NB medium could help the formation of embryogenic callus;nutrient matters could increase callus induction rate,but at the sacrifice of E.C.rate.According to the callus growth curves,the proper time for subculture was between 24 to 33 days.
