蚕豆（Vicia　faba　L．）叶肉细胞中ABA的胶体金免疫电镜定位

Immunogold Electron-Microscopy Localization of Abscisic Acid to Mesophyll Cells of Vicia faba L.

利用胶体金免疫电镜定位技术对蚕豆叶肉细胞中ＡＢＡ定位的研究表明，在以ＡＢＡ抗体处理的切片中，叶绿体有大量的金颗粒标记，细胞质和细胞核也有金颗粒标记，但液泡和细胞壁中没有金颗粒标记。免疫染色前用胰蛋白酶处理可显著增强金颗粒标记密度，而不用ＥＤＣ固定或以免疫前兔血清处理的切片中几乎没有金颗粒标记。本实验为蚕豆叶肉细胞中ＡＢＡ的分布提供了直接的证据并说明了该技术是研究ＡＢＡ定位的一种可靠的方法。

With immunogold electron microscopy the study on the localization o f abscisic acid (ABA) in mesophyll cell of Vicia faba has been undertaken.The results showed that, in thin sections treated with ABA antibody,chloroplasts, cytosolic comportments and the nuclei were labelled by gold particles, but chloroplasts were labelled by more gold particles. However, no gold particles were found in cell wall and vacuoles. Treating the sections with trypsin before immunostuining could increase the gold particle density substantially. The sections with no EDC fixation or treated with preimmune rabbit serum were devoid of gold particle label. this study has provided direct evidences for the localization of ABA in mesophyll cells of Vicia faba and shows that the technique described in this papar is a reliable method for the study of ABA localiztion.
