白念珠菌对角质形成细胞Toll样受体2表达的影响 被引量:2

摘要 目的:探讨白念珠菌和甘露聚糖对人角质形成细胞Toll样受体2(TLR2)表达的影响。方法:用白念活菌和甘露聚糖分别刺激培养的人角质形成细胞24h,然后用逆转录聚合酶链反应(RTPCR)检测细胞TLR2mRNA表达,免疫组化SP法检测TLR2蛋白表达,并进行半定量分析。结果:正常培养的人角质形成细胞有TLR2mRNA和蛋白表达,白念珠菌和甘露聚糖刺激角质形成细胞24h后,TLR2表达量没有统计学差异(P>0.05)。结论:角质形成细胞有TLR2mRNA和蛋白水平的组成性表达,白念珠菌和甘露聚糖对TLRR2表达均无明显影响,推测角质形成细胞表面可能存在多个识别念珠菌及其产物甘露聚糖的受体,TLR2只是其中之一。 Objective: To investigate the influence of Candida albicans and mannan on the expression of Tolllike receptor 2 (TLR2) in cultured human keratinocytes. Methods: The keratinocytes were stimulated with Candida albicans and mannan for 24 hs and the expression of TLR2 mRNA was measured by RT- PCR. TLR2 protein was stained by immunohistoohemistry technique. Semi - quantitative analysis was made for the expression of TLR2 mRNA and TLR2 protein. Results: TLR2 mRNA and TLR2 protein were detectable in keratinocytes. There was no statistically significant difference between keratinocytes stimulated with Candida albicans and mannan and those not stimulated cells for TLR2 mRNA and TLR2 protein ( P > 0.05). Conclusion: Keratinocytes can constituently express TLR2 mRNA and TLR2 protein. Neither Candida albicans nor mannan may regulate the expression of TLR2 in cultured keratinocytes. Our findings suggest that there are several receptors recognizing Candida albicans and mannan on the surface of keratinocytes and TLR2 may be one of them.
