摘要
目的采用生物信息学分析技术比较葡萄胎和绒毛膜癌基因表达谱,筛选与绒毛膜癌恶性演进相关的新基因。方法对两组葡萄胎和绒毛膜癌基因表达谱的共表达基因数据进行分层聚类分析(hierarchicalcluster),找到有相似表达趋势的候选基因,再进行电子Northern分析及基因结构功能初步分析,用半定量RT PCR技术对3个筛选基因进行初步鉴定。结果在所有绒毛膜癌基因表达数据库中找到共表达候选基因52条,经聚类分析初步筛选出绒毛膜癌相关基因19条,主要包括发动蛋白(dynamin,L07807)、锌指蛋白184(zincfingerprotein184,U6656)、钙调蛋白(calmodulin,U12022)、羧肽酶M(car boxypeptidaseM,BC022276)、剑蛋白p60(ka taninp60,AF056022)、神经钙蛋白结合蛋白1(calcineurinbindingproteincabin1,NM_012295)和转导蛋白样增强子蛋白1(transdu cin likeenhancerprotein1,M99435)等。结论通过生物信息学分析技术,初步筛选了绒毛膜癌相关基因,为寻找绒毛膜癌发生与演进的关键基因提供了新的依据。
OBJECTIVE: To analyze the data of gene expression profiles of choriocarcinomas and complete hydatidiform moles (CHM) and to screen novel genes that are associated with tumor progression in choriocarcinomas. METHODS:The candidate genes with similar expression tendency were found by hierarchical clustering the data of gene expression profiles of choriocarcinomas and CHM. The gene function, structure and expression in normal and neoplastic tissues were analyzed by electric northern and other bioinformatic tools. Three selected genes were analyzed by β-actin semiquantitative reverse transcriptase-PCR to confirm the data. RESULTS: A total of 52 genes were identified for candidates for coexpression in all cases of gene expression profiles of choricarcinomas, of which 19 genes were selected for the choricarcinoma-associated genes by hierarchical clustering, such as dynamin (L07807), zinc finger protein 184 (U6656), calmodulin (U12022), katanin p60(AF056022), carboxypeptidase M (BC022276), calcineurin binding protein cabin 1(NM_012295) and transducin-like enhancer protein 1(M99435), etc. CONCLUSION:The preliminary identification of choriocarcinoma-related genes by analysis of mass data of gene expression profiles provides new basis and information for seeking key genes in relation to progression and metastasis of choriocarcinoma.
