摘要
目的:探讨评价细胞毒性的方法。方法:分别采用MTT法、MTS法和传统的血球计数法比较医用高分子材料的浸提液对小鼠成纤维细胞(L - 92 9)相对增殖率的影响。结果:采用3种测定方法测定此类医用高分子材料,均呈现出高的细胞相对增殖率,其细胞毒性为1级,即无细胞毒性。结论:在常规细胞毒性测试法的基础上采用MTT法和MTS法,能更快捷准确地测定细胞的增殖反应。MTS法与MTT法相比较,其灵敏度更高,操作更简便。
Objective: To probe the evalu a tion on approaches to cytotoxicity. Methods: The relativ e proliferation rate of L?929 mouse fibroblasts was determined to compare the e ffect of the extract of polymeric material for medical use by MTT?assay, MTS?a ssay and the traditional method of blood cell counts. Results: The results indicated that the relative growth rate induced was high by the three approaches of measurements, and the cytotoxicity was grade I,meaning t hat the material had no cytotoxicity. Conclusion: On the basis of routine cytotoxicity test, the application of MTT?assay and MTS?assa y could determine the cell propagation reaction faster and more accurately,and MTS?assay proved to have higher sentitivity and thus to be more convenient an o peration than MTT?assay.
