摘要 目的 研究乙醛对肝星状细胞(hepaticstellatecell,HSC)增殖、细胞周期、细胞周期蛋白cyclinD1及细胞外信号调节激酶(Extracellularsignal-regulatedkinase,ERK)磷酸化蛋白表达的影响。方法 体外培养HSC株;以MTT法检测细胞增殖,流式细胞仪检测细胞周期,RT -PCR方法检测HSC内CyclinD1mRNA表达,Westernblot检测乙醛刺激后HSC内ERK活性的变化。结果 HSC被乙醛刺激后,细胞增殖、细胞由G1→S期转化增多、细胞周期蛋白Cy clinD1mRNA表达增加、磷酸化ERK(p -ERK)水平增高。与空白对照组比较,统计学分析有显著差异(P <0 .0 5 )。结论 乙醛刺激HSC后引起HSC增殖与ERK信号传导通路活化有关。 Objective To investigate the effect of acetaldehyde o n the proliferation of rat hepatic stell ate cells.Methods Rat hepatic stella te cells were cultured in vitro,HSC were collected,the changes of the proliferat ion,the changes and expression of Cyclin D1 mRNA,and the level of the p-ERK were detected respectively by means of MTT,fl ow cytometry,RT-PCR,western blot.Results The cells prolifer ation,p-ERK level,and CyclinD1 mRNA expr ession were enhanced apparently after HS C were stimulated by acetaldehyde;the tr ansition from G1 to S phase was also acc elerated in HSC stimulated by acetaldehy de;there existed a significant statistical difference between experimen tal group and control group (P<0.05).Conclusion ERK could be ac tivated significantly after HSC were sti mulated by acetaldehyde,and HSC prolifer ation stimulated by acetaldehyde is rela ted to ERK signal transduction pathway.
