肝、肺、胰腺不同组织对胃癌GASL细胞侵袭的趋向引导作用

摘要 目的研究裸鼠肝、肺、胰腺不同组织对胃癌GASL细胞系侵袭能力的影响,同时观察胃癌细胞在各种不同组织中的生长和粘附情况。方法利用Transwell小室侵袭实验,分别比较从10只BALB/C裸小鼠体内提取肝、肺、胰腺组织诱导人胃癌细胞系GASL移动侵袭能力。结果无论培养液中是否含有血清,癌细胞对肝组织的粘附率都高于肺组织和胰腺组织者。在元血清条件下,接种于塑料板上的癌细胞增殖停滞并死亡,而接种在不同组织上的癌细胞能继续生存和增殖,并以肝组织上者占优势。通过Transwell小室法癌细胞穿过肝、肺、胰腺组织诱导GASL细胞移动侵袭的细胞数分别是(13.1±0.9)、(5.7±0.3)、(11.2±0.4),与肺组织相比,肝组织和胰腺组织具有较明显诱导GASL细胞的移动能力,差异有显著性(P<0.01)。结论(1)不同组织微环境能为肿瘤细胞的生长提供有利的条件,具有补充元血清条件下缺少营养物质和生长因子的作用;(2)不同组织微环境决定了GASL细胞系的侵袭表型。 Objective To study the effect of different tissues of liver, lung and pancreas of nude mice on the invasion of human gastric carcinoma cells GASL, meantime investigated the growth and adhesion property of a gastric carcinoma cell in vitro on the different tissues. Methods The Transwell in vitro invasion assay was used to compare the ability of migration and invasion of human gastric carcinoma cells GASL, inducing by extracts of liver, lung, pancreas from ten nude mice. Results Either in SFM or SSM, the carcinoma cells showed a higher coefficient of adhesion to the liver tissue than to the lung and pancreas tissue, in SFM, cells pkted on plates ceased proliferation and died, whereas those on the different tissues, particularly the cells on the liver tissue survived and proliferated. Transwell invasion assay showed the numbers of cells in response to liver, lung and pancreas tissue were (13.1###0.9), (5.7###0.9), (5.7###0.3) and (11.2###0.4), respectively. In comparison of lung tissue, the liver and pancreas tissue showed significant differences to promote the migratory and invasive ability for GASL (P<0.01). Conclusion (1)The microenviroment of different tissue may provide conditions favourable for cell growth, and may compensate in effect of the deficiency of nutrition and growth factors when serum is absent in the culture medium; (2)The tissue enviroment may have definitive effects on the tumor cells phenotypes.
