摘要 目的 检测宫颈鳞癌中KAI1在mRNA水平的表达,探讨其与宫颈癌侵袭转移的关系。方法 采用逆转录-多聚酶链反应(RT-PCR)技术检测新鲜组织KAI1 mRNA的表达,宫颈鳞癌组52例,正常对照组21例。结果 (1) KAI1 mRNA 在正常宫颈上皮及宫颈癌组织中阳性率分别为100%,61.53%,差异具有显著意义(P<0.05)。(2) 在高、中、低分化宫颈癌组织中KAI1 mRNA阳性率分别为76.47%、68.18%和30.70%,三组间差异有极显著意义(P<0.01)。(3) KAI1 mRNA在≤ⅡA、ⅡB、Ⅲ/Ⅳ期宫颈癌组织中表达阳性率分别为68.00%、55.56%和55.56%,阳性率在各期间差异无显著意义(P>0.05)。(4) KAI1 在无淋巴转移组的阳性率(27/34,79.41%)明显高于有淋巴转移组(5/18,27.78%),差异有显著意义(P<0.05)。结论 KAI1 mRNA在宫颈鳞癌组织中表达减少,其表达与细胞分化及转移有关,提示KAI1可能参与了宫颈癌的发生发展,可能在一定程度上具有预测宫颈癌侵袭转移的潜能。 Objective To study the expression of KAI1 mRNA in squamous cell carcinoma of cervix and explore the relationship between KAI1 and invasiveness and metastasis of cervical cancer.Methods 73 cases of fresh freezing tissues were detected by RT-PCR, including 21 cases of normal cervix and 52 cases of tumor tissue of primary cervical squamous. Results The positive rate of KAI1 mRNA in normal cervix and cervical carcinoma were respectively 100% and 61.53%, there was a significant difference between them(P<0.05). The positive rate of KAI1 mRNA in high, moderate and poor differentiated group in cervical cancer were respectively 76.47%、68.18% and 30.70%, there weresignificant difference samong three groups(P<0.01). The positive rate of KAI1 mRNA in stage ≤ⅡA, stage ⅡB and stage Ⅲ/Ⅳ were respectively 68.00%、55.56% and 55.56%, there was no significant difference among three groups(P>0.05). The positive rate in non-metastasis and metastasis group of cervical cancer were respectively 79.41% (27/34) and 27.78%(5/18), there was a significant difference between them(P<0.05). Conclusion There was a down-regulation of KAI1 in cervical cancer. The expression of KAI1 mRNA is related with the differentiation and metastasis of cervical carcinoma. KAI1 might play an important role in the progression of cervical cancer. Its detection may predict the invasive and metastatic potential of cervical carcinoma.
