摘要 目的 采用SPA法在镀金电极表面进行抗体固定,探索压电免疫传感器再生使用的最佳条件。方法 对压电免疫传感器的检测时间、特异性,及再生试验的最佳试剂、时间进行实验研究。结果 构建的压电免疫传感器检测时间为15min,无交叉反应;NaOH溶液再生10次后,频率响应下降为原有的75%。结论 所构建的压电免疫传感器检测时间短,特异性好,可用于临床标本的检测,NaOH溶液用于石英晶体再生的效果较好,极大地降低了使用成本。 OBJECTIVE To study the best condition about regeneration ability of piezoelectric immunosensor which immobilized anti-human antibody by SPA. METHODS We did experiments to find the detection time of the immunosensor and the best reagent and concentration to regenerate the used crystals. RESULTS The piezoelectric quartz crystal immunoglobulin microarray immunosensor could complete the detection in 15min without nonspecific response. More than 75% of the original response of the quartz crystals was maintained after 10 assays which used 1.2M NaOH as regeneration reagent. CONCLUSIONS The results could be completed in short time and without nonspecific response, which showed that the microarray immunosensor could be used for clinical detection. Using 1.2M NaOH as regeneration reagent gives the better result, which reduced the cost largely.
