摘要 肿瘤的单纯基因治疗或单纯肿瘤特异性增殖病毒治疗,都取得了一定的疗效,但都无重大突破。本文提出一种癌症的靶向基因病毒治疗新策略(targetinggene virotherapyofcancer),即将基因治疗与病毒治疗各自的优点结合起来,取得的疗效比上述任何单一的治疗都好。实验所用的载体是改造过的腺病毒ZD55或hTERT Adv,但单用一个基因还不足以完全消灭实验动物的肿瘤,因此进一步采用双基因治疗方案,即targetingdualgene virotherapyofcancer。如果两个基因选择恰当,当它们有协同效应或互补作用时,如Trail与K5,Trail与Smac,则基本可以全部消灭实验动物所荷肿瘤。此外,要取得癌症治疗的好结果,靶向性也很重要,故一种新的双靶向病毒调控双基因的癌症治疗策略被创造出来,并取得了很好的效果,申请了专利。无肠腺病毒为第三代腺病毒载体,它容量大,可克隆入很大的基因,又无免疫原性,可长期使用,如将前面各种良好肿瘤治疗策略结合运用到无肠腺病毒载体中,一定会构建出最好的肿瘤治疗方案,会使肿瘤治疗取得极好的效果。
