草木樨状黄芪单细胞培养再生植株 被引量:4

摘要 对草木樨状黄芪茎段愈伤组织,单细胞采取振荡悬浮和静止浅层培养的结果表明,振荡培养优于静止浅层培养,振荡培养条件下单细胞2~3天进行第一次分裂,10天左右形成小细胞团,进而发育为胚状体或愈伤组织,30天左右小愈伤组织直径达1~2mm,愈伤组织和胚状体再生频率为88块/ml(0.2%),是静止培养的3.8倍。经继续振荡培养,小愈伤组织和胚状体即可分化或萌发出大量幼苗。小愈伤组织经固体培养基增殖培养后,转入分化培养基可100%分化,目前已获得大量再生植株。 Monocells were isolated from the suspension cultures which originated from calli derived from stem sebments of sterile seedlings of Aslragalus melilotoides. The monocells were cultured in modified MS liquid media by two methods.One was vibrational culture in culture bottle ,and the other was motionless culture in petri dish.By comparison ,the vibrational culture was a better method.By which, the regeneration frequency of mi- crocallus was 88 pieses/ml(0.2%)which was 3.8 times as much as that by motionless culture,The differentiation frequenies of calli cultured in original liquid media or trans- ferred to solid differention media were all 100% Up to now,a large number of regen- eration plants were obtained.
