摘要
目的:探讨人巨细胞病毒(human cytomegalovirus HCMV)感染对人早孕绒毛及蜕膜组织的内分泌影响.方法:采用体外培养早孕绒毛及蜕膜组织技术,给予HCMV感染建立体外HCMV感染早孕绒毛及蜕膜模型,采用放射免疫法检测HCMV感染后早孕绒毛及蜕膜组织培养上清液中hCG和PRL的水平.结果:①正常早孕绒毛培养液中hCG分泌峰值在培养后第7天.给予高浓度HCMV干预后,hCG水平显著下降(P<0.01),给予中浓度HCMV干预后,hCG水平明显下降(P<0.05),给予低度HCMV干预后,hCG水平于培养的第1~3天有所下降,与正常对照组无显著差别(P>0.05).②正常蜕膜培养液中PRL分泌峰值在培养后第5天达峰值,给予高浓度HCMV干预后,PRL水平显著下降(P<0.001),给予中浓度HCMV干预后,PRL水平明显下降(P<0.01),给予低度HCMV干预后,PRL水平于培养的第1~5天明显下降(P<0.05)后渐恢复至正常水平.结论:HCMV感染可影响早孕绒毛及蜕膜组织的hCG和PRL的分泌,导致不良妊娠结局.
Objective:To explore the effect of human cytomegalovirus (HCMV) infection on the endocrine function of human chorionic trophoblast cells and decidual cells in the first trimester placenta explants.Methods:In vitro HCMV infection human chorionic trophoblast and decidual model were built with in vitro culture technique. Human chorionic gonadotropin (hCG) and chorionic tissues and prolactin (PRL) levels in chorionic trophoblast and decidual cells after HCMV infection were determined by radioimmunoassay (RIA).Results:① The hCG secretive peak in normal culture appeared at the 7th day. The hCG level significantly dereased ( P<0.01, P <0.05) after intervention with high and moderated concentration of HCMV, while decreased from 1st to 3rd day after intervention with low concentration of HCMV ( P >0.05). ②The PRL secretive peak in normal culture appeared at the 5th day. The PRL level significantly dereased ( P<0.001, P <0.01) after intervention with high and moderated concentration of HCMV, while decreased from 1st to 5th day ( P <0.05) after intervention and later up to normal with low concentration of HCMV.Conclusion:HCMV infection could result in poor pregnant outcome by means of the effect on endocrine function of the first trimester placenta.
