川芎嗪对类风湿性关节炎患者外周血单个核细胞IL-12mRNA表达的影响

Effects of Ligustrazin on the expression of IL-12mRNA in periphe ral blood mononuclear cells of patients with rheumatoid arthritis

目的:探讨川芎嗪对培养的类风湿性关节炎(RA)活动期、静止期外周血单个核细胞(PBMC) IL- 12 m RNA表达的影响。方法:应用细胞培养技术,采用RT- PCR方法,检测细胞培养液IL - 12 m RNA表达水平。结果:RA患者活动期、静止期IL - 12高度表达(P <0 .0 5 ) ,川芎嗪可以抑制RA活动期PBMC IL - 12合成,并对活动期PBMCIL - 12 m RNA有抑制作用(P<0 .0 5 )。结论:川芎嗪可通过抑制RA患者中PBMCIL - 12 m RNA的表达,减轻RA进展。

Objective: Regulatory effects of Ligustrazin on the e xp ression of IL-12mRNA in peripheral blood mononuclear cells (PBMC) from the patie nts with rheumatoid arthritis (RA) were detected. Methods: The cel l cultured technique was used and ,the IL-12mRNA level was assayed by RT-PCR. Results: The IL-12 level was higher in the active and acute stages of RA patients than that of the normal controls. Ligustrazin suppressed the IL- 12 synthesis and the IL-12mRNA expression (P<0.05) . Conclusion: Ligustrazin down-regulate the IL-12mRNA expression of RA patie nts, thus contributing to amelioate the condition of RA patients.
