摘要
目的 探讨人肝癌多药耐药细胞株survivin蛋白表达与肝癌细胞多药耐药的关系。方法 用阿霉素(DOX)浓度梯度递增诱导法,建立人肝癌多药耐药细胞株(Bel- 74 0 2 / DOX)。用流式细胞仪检测肝癌细胞凋亡率,同时用蛋白印迹(Western blot)检测其survivin蛋白表达。结果 随着培养液中DOX浓度的增加,Bel- 74 0 2 /DOX的凋亡率逐渐增加,survivin蛋白的表达逐渐增强。结论 Survivin蛋白的表达变化可能与人肝癌细胞获得性耐药有关。
Objective To explore the expression of survivin protein in multi drug-resistant human hepatocellular carcinoma cell line.Methods The doxorubicin-resistant human hepatocellular carcinoma cell line (Bel-7402/D OX) was established in vitro by gradual increase of doxorubicin concentrations i n culture.The apoptosis rates of the hepatocellular carcinoma cell were m easured by flow cytom eter (FCM),and the survivin protein were detected by Western blot.Results With progressively increasing the concentration of doxorubicin,the apoptosis rates an d the expression level of survivin protein increased gradually.Conclusions The expression of survivin protein might co rrelate with the inquired drug-resistance of human hepatocellular carcinoma.
