HPLC法测定双黄连胶囊中连翘酯苷A及连翘苷的含量

Determination the Content of Forsythoside A and Forsythin in Shuanghuanglian Capsules by HPLC

目的:建立中成药双黄连胶囊中连翘酯苷A及连翘苷的含量测定方法。方法:采用Phenomenex-ODS-C18(250 mm ×4.6 mm,5 um)柱,以甲醇(含1%的四氢呋喃)-水(含0.01 mol·L-1磷酸二氢钾,PH 3.2)为流动相进行梯度洗脱,检测波长280 nm。结果:连翘酯苷A及连翘A的线性范围分别为7.8-139.5μg·ml-1和2.6～47.7μg·ml-1,其线性关系良好。连翘酯苷A平均回收率为100.3%,RSD为0.75%;连翘苷平均回收率为99.2%,RSD为1.03%。结论:该方法准确可靠,可用于本制剂的质量控制。

Objective: To establish a HPLC method for determination of the contents of forsythoside A and forsythin in Shuang-huanglian capsules. Method: A Phenomenx-ODS- C18(250 mm×4.6 mm,5 um)was used. The mobile phase was methanol ( containing 1% THF) water (0.01 mol·L-1 potassium dihydrogen phosphate solution, pH3.2) as gradient elution. the detecting wavelength was at 280 nm. Result: The linear ranges of forsythoside A and forsytin were at 7. 8-139. 5 μm·ml-1 and 2. 6-47. 7 μm·ml-1, respectively, the both showed a good linear correlation. the average recoveries were respactively 100. 3% ( Forsy thoside A RSD 0. 75% ) and 99.2% (Forsythin, RSD 1.0% ). Conclusion: The method is reliable, accurate, and very suitable to the quality control of Shuang-huanglian capsules.
