摘要
研究了翻曲、不同的培养时间及。NaCl对分离得到的毛霉的蛋白酶活力的影响,探讨了不同的毛霉蛋白酶提取方法,并对比了福林酚法测定蛋白酶活力时的最佳吸收波长。结果表明:该株毛霉麸曲固态培养72h时蛋白酶活力最高;培养24h后翻曲1次能显著提高酶活力;用盐水麸曲制曲培养时,用蒸馏水提取可获得较高活力的复合蛋白酶;测定蛋白酶活力时,最佳吸收波长为690nm。
This paper dealt with the effect of culture disturbing, culture time, and NaCl concertration on protease activity of the mucor spp. Several extraction methods of mucor protease and optimal absorbing wavelength for detection of protease acticity were also studied. The results were that high protease activity could be obtained with tsolid-medium cultivation for 72h. One culture-disturbing after 24h cultivation could remarkablu increase protease activity, and extraction with distilled water during koji cultivation could obtain compound protease with high activity. The optimal absorbing wavelength was 690nm。
