摘要
采用松花粉垂钓法分离到一株Docosahexaenoicacid(DHA)高产菌FJU 512.该菌株DHA含量高(占总脂肪酸的56. 24 % ),其它长链杂酸含量少(仅有docosapentaenoicacid, DPA),极具开发应用价值.高密度培养可获得33gL-1生物量.该菌株行二分裂生长,没有分生胞子.对其18SrRNA基因进行了克隆测序并登录GenBank(AY758384).依据18SrRNA基因建立的系统进化树表明:该菌与Schizochytriumlimacinum具有紧密的亲源关系.
A strain FJU-512 highly producing docosahexaenoic acid (DHA) was isolated by using pine pollen as bait. Its single cell oil was very valuable since its polyunsaturated fatty acid contained only little docosapentaenoic acid but much DHA, which was up to 56.24% in the total fatty acids. The strain biomass of 33 g L -1 was obtained by high-density culture. Its growth was dependent on binary division without spores. Its 18S rRNA gene was successfully cloned by PCR and sequenced (GeneBank: AY758384). The phylogenetic tree based on 18S rRNA gene showed that Schizochytrium sp. FJU-512 was close to Schizochytrium limacinum . Fig 7, Tab 2, Ref 29
