Rapid detection of influenza virus by immunofluorescent staining of shell vial cultures

摘要 目的:为在流感样疫情的调查中能够快速准确地查清流感病原,提高公共卫生突发性应急事件的快速反应能力。方法:摇床吸附微量细胞板短期培养法的荧光抗体染色用于快速检测流感病毒并与常规的病毒分离方法作比较。结果:对83份流感疑似病人含漱液的快速检测结果与常规的细胞培养法病毒分离结果相比较,在病毒分离阳性的45份标本中,快速检测法也有34份阳性,阳性符合率为75%,敏感性测定达到了能够检测出0 01个血凝单位浓度的流感病毒。结论:微量细胞板荧光染色法检测流感病毒的敏感性要优于一般市售的快速诊断试剂盒。检测时间约在24h内,达到了快速准确的目的。 Objective:To rapidly determine the pathogen of influenza during outbreak; increase rapid response faculty of public health authority in counterpunch of paroxysmal epidemic situation.Methods:The immunofluorescent staining of shell-vial cultures was used for rapid detecting influenza virus , and compared with conventional viral isolation.Results:The samples of 83 patients were measured with these 2 methods.In 45 samples of influenza virus positive measured by viral isolation,34 samples were also positive measured by immunofluorescence staining of shell-vial cultures, the positive coincidence rate was 75%.The sensitivity had also reached and could measure out 0.01 unit′s hemagglutinin of influenza virus .Conclusion:The immunofluorescent staining of shell-vial cultures was better than other methods with commercially available rapid diagnose kits.
