转化生长因子β_1在扩张型心肌病大鼠心肌纤维化中的作用 被引量:3

摘要 目的探讨转化生长因子(TGF)β1在扩张型心肌病大鼠左室心肌纤维化过程中的作用。方法用呋喃唑酮饲养Wistar大鼠建立扩张型心肌病大鼠模型,超声心动图检测大鼠心功能,病理学测量大鼠左室内径和左室游离壁厚度,HE和VG染色分别观察大鼠心肌细胞和间质胶原纤维的变化,免疫组化检测左室心肌组织TGFβ1表达水平。结果扩张型心肌病大鼠左心室内径增大、游离壁变薄,左室收缩功能下降;心肌细胞肥大、变性,核增大、分裂、畸形;间质胶原纤维明显增生;左室心肌组织TGFβ1表达水平明显增高。结论TGFβ1在扩张型心肌病大鼠左室心肌纤维化中起重要作用。 AIM: To study the action of transforming growth factor (TGF) β_1 in rats myofibrosis with furazolidone induced dilated cardiomyopathy. METHODS: The Wistar rat model of dilated cardiomyopathy was created by fed with Furazolidone .The cardiac funtion of rats was detected by echocardiography. Left ventricular interior diameter and the thickness of ventricular free wall were measured afrer the rats were killed. Myocardial cell morphology and interstitial collagen were observed by HE and V-G stain. TGF β_1 was detected by immunohistochemical method. RESULTS: The left ventricular interior diameter became wide and the thickness of ventricular free wall was thin. The left ventricular contract function decreased significantly. Cardiac myocyte hypertrophy and lysis accompanied with nucleus augmentation, differetiation and aberration. The level of TGF β_1 was increasesd. All were seen in the rat with dilated cardiomyopathy. CONCLUSION: TGF β_1 is the main role of left ventricular myofibrosis in rats with furazolidone-induced dilated cardiomyopathy.
