锌对镉所致大鼠含锌酶活性降低的保护作用 被引量:4

The Protective Effects of Zinc Pre-supplement on Decreasing the Activities of ALP and SOD in Liver and Serum of Rats,Treated with Cadmium

摘要 [目的]探讨锌对镉所致大鼠肝脏及血清中含锌酶[主要为碱性磷酸酶(ALP)、过氧化物歧化酶(SOD) ]活性降低的保护作用。[方法]将3 6只Wistar大鼠随机分成6组:(1)正常对照组;(2 )补锌对照组;(3 )低镉组(0 .5mg/kg) ;(4 )锌+低镉组;(5 )高镉组(2mg/kg) ;(6)锌+高镉组。均以普通饲料饲养。(1)、(3 )、(5 )组自由饮去离子水,(2 ) (4 )、(6)组自由饮加锌的去离子水后第6天,(3 )、(4 )组和(5 )、(6)组分别腹腔注射0 .5、2mg/kgCd离子折算量的氯化镉溶液,(1)、(2 )组腹腔注射生理盐水,均2 4h后断头处死,取出肝脏,分别测定各组肝脏及血清中ALP、SOD活性。[结果]正常对照组肝组织SOD活性为(77.963±12 .915 )NU/mg蛋白,ALP活性为(13 .0 66±4.198)U/L ,高镉组SOD活性降至(60 .0 2 1±9.481)NU/mg蛋白(P <0 .0 5 ) ,ALP活性降至(1.93 9±1.0 5 6)U/L(P <0 .0 5 )。对照组血清ALP活性为(3 1.2 60±2 .45 9)U/L ,染镉后降至(17.677±3 .70 4)U/L(P <0 .0 5 ) ,预先补锌后再染镉可见锌+高镉组肝组织SOD活性升高(P <0 .0 5 ) ,锌+低镉组血清ALP活性升高(P <0 .0 5 )。[结论]锌对镉所致大鼠肝脏及血清中含锌酶活性的降低有保护作用。 Objective] To explore the protective effects of zinc pre-supplement on decreasing the activities of alkaline phosphatase(ALP)and superoxide dismutase(SOD)in liver and serum of rats treated with cadmium. [Methods] Thirty-six Wistar rats were randomly divided into six groups:blank control group,zinc pre-supplement control group,cadmium low dose group(0.5 mgCd/kg),low dose cadmium with zinc pre-supplement group,cadmium high dose group(2 mgCd/kg) and high dose cadmium with zinc pre-supplement group.Every group was given common feed.Zinc pre-supplement group was given additional zinc for 6 days.Then the activities of ALP and SOD were determined 24 hours after cadmium was injected intraperitoneally at the different doses. [Results] The activities of SOD in liver in control group is 77.963±12.915 NU/mg Pr and it decreased to(60.021±9.481) NU/mg Pr in cadmium high dose group(P<0.05).The activities of ALP in liver in control group is(13.066±4.198) King Unit/L and it decreased to 1.939±1.056 King Unit/L in cadmium high dose group(P<0.05).The activities of ALP in serum in control group is(31.260±2.459) King Unit/L and it decreased to (17.677±3.704) King Unit/L in cadmium high dose group(P<0.05).In the groups of zinc pre-supplement and cadmium injection SOD and ALP activities increased in different degree. [Conclusion] Cadmium could inhibit the activities of ALP and SOD in liver,and the activities of ALP in serum respectively.Zinc pre-supplement to rats could antagonize these Cd effects in certain degree.
