摘要
对PRRSV诱导Marc145细胞凋亡的动态变化规律进行了研究,实验收集PRRSV SC1株感染Marc145细胞后0、24、48、72、96、120、144h和同一时间点未接毒的细胞样品,用DNALadder定性检测和ELISA定量检测细胞凋亡的变化规律。定性检测结果表明,PRRSV能诱导Marc145细胞发生凋亡,病毒感染后凋亡细胞DNA电泳呈典型“梯状”条带。定量检测结果表明,Marc145细胞在PRRSV感染后24h开始出现凋亡,48h凋亡更加明显,72h达到高峰,明显高于对照组。
For researching dynamic changing rules of apoptosis in PRRSV-induced Marc 145 cell line, samples of Marc 145 cell line infected with PRRSV at 0, 24, 48, 72, 96, 120, 144?h and corresponding time points control Marc 145 cell line were collected, qualitative and quantitative apoptosis of cell cultures were detected by DNA Ladder and ELISA separately. The result of DNA Ladder indicates that PRRSV can induce apoptosis of Marc 145 cell line in vitro and the DNA of cell cultures presented classic Laddering pattern. The result of ELISA indicates that Marc 145 cell begins to show apoptosis at 24?h post-infected, apoptosis changes obviously at 48?h and the peak value of apoptosis appeares at 72?h , higher than that of control group.
