摘要 目的:研究FK506促进周围神经再生的作用机制。方法:将人胚坐骨神经植块分4组:0.1μg/ml、1μg/ml和10μg/ml的FK506药物组以及空白对照组,比较各组雪旺细胞从植块中迁出的时间、96 h迁移范围和MTT活性。结果:FK506 药物组人胚雪旺细胞较早从植块中迁移出来,96h迁移范围明显大于对照组,但不同药物浓度组之间无明显差异。结论:FK506 能促进体外培养的人胚雪旺细胞迁移并抑制成纤维细胞生长,这种作用在较低浓度时也能产生明显效果。 Objective: To explore the mechanism of FK506 in promoting peripheral nerve regeneration. Methods: Explants from human embryo sciatic nerves were randomly divided into 4 groups,the control group and FK506 groups with different doses of 0.1μg/ml, 1μg/ml and 10μg/ml.Results: Human fetal Schwann cells migrated out of the explants earlier and occupied larger range in 96h, and higher MTT activity than that of the control group, but there were no significant differences among the 3 different doses.Conclusion:FK506 could promote Human fetal Schwann cell migration in vitro and restain the growth of fibroblast. This effect of FK506 was evident in low dose level.
