摘要
目的用健康兔做在体心房室结缺血再灌注损伤模型,观察房室结缺血再灌注损伤的超微结构变化及BCL-2基因蛋白表达。方法健康兔30只,随机分为3组,A组(假手术组)n=10,B组(缺血30min,再灌注30min)n=10,C组(缺血30min,再灌注240min)n=10。结果A组血清天冬酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、肌酸肌酶(CK)未升高,免疫组化检测房室结BCL-2基因蛋白未见表达,超微结构无变化。B、C组AST、LDH、CK及BCL-2基因蛋白表达逐渐升高,超微结构中细胞水肿逐渐明显,线粒体肿胀、嵴断裂逐渐明显,肌丝逐渐收缩,细胞核染色质集块边聚明显。结论心房室结缺血再灌注损伤随再灌注时间延长变化明显。
Objective The animal models of rabbit ischemia-reperfusion atrioventricular node were used to study the changes of ultrastructure and expression of BCL-2 protein. Methods In this experiment, 30 rabbits were divided into 3 groups.A group(normal group)n=10,B group(ischenia 30min,re- perfusion 30min)n=10,C group(ischenia 30 min,reperfusion 240min)n=10. Results All of AST? LDH? CK,espression of genic protein and ultractrucure were normal in A group;Both AST LDH CK and expression of genic protein were higher and higher of B to C group,In ultracture the cellular were smollen,the mitochondria were swollen,the cristae were disorganized,the flesh fibers were shrinked,the chromatin of nucleus were massed and margined were more and more of B to C group. Conclusion The change of ischemia-reperfusion injury groups were increased in a time-dependent manner of reperfusion.
