摘要
目的:建立一种蝉拟青霉多糖的含量测定方法。方法:应用分光光度法测定蝉拟青霉多糖的含量。结果:测定波长485 nm,在0.005-0.230 mg·ml-1范围内多糖浓度与吸收度有良好的线形关系,回归方程A=8.130C+0.033,r2= 0.996,平均回收率为97.99%±3.96%,RSD=4.04%。蝉拟青霉菌丝体的多糖含量为1.92%。结论:该方法操作简便,重复性好,显色稳定,可用于蝉拟青霉多糖的含量测定。
Objective: To find a method for detecting the polysaccharlde from Paecilomyces cicadae. Methods: The polysaccharide content was detected by the spectrophotometry. Results:Detected at 485 nm wavelength,the linear regressive equation was A = 8. 130C + 0. O33,r2 = 0. 996 of which linear range was 0. 005-0. 230 mg·ml-1. The average recovery was 97. 99%±3. 96%(RSD = 4. 04%). The polysaccharide content of mycelial paecilomyces cicadae was 1. 92%. Conclusion: The method is convenient and stable, and can be used for detecting the polysaccharlde from paecilomyces cicadae.
