摘要
目的:观察右旋柠烯(d limonene ,DL)对大鼠乳腺增生及血雌激素和孕激素水平的影响。方法:苯甲酸雌二醇-黄体酮注射复制乳腺增生模型;HE染色观测乳腺组织学改变;放射免疫法检测血雌激素和孕激素水平。结果:右旋柠烯低、中、高剂量均能减少大鼠乳腺小叶数(9.2 0±2 .3 2、6.90±1.2 0、6.10±1.3 7)、小叶腺泡数(8.2 0±1.75、6.10±2 .2 3、5 .10±1.5 2 )和导管上皮层数(5 .3 0±0 .95、4.5 0±1.2 7、3 .10±0 .99) ,与模型组(分别为10 .0 0±1.5 4、12 .91±3 .88和6.17±1.5 9)比较有显著性差异(P <0 .0 1)。右旋柠烯高、中剂量亦能降低血雌激素水平(2 8.3 5±2 .16、2 5 .17±1.2 5 ) ,提高孕激素水平(3 3 .84±2 .2 3、3 5 .3 9±2 .2 0 ) ,与模型组(分别为3 2 .63±0 .93 ,3 1.77±1.48)比较差异显著(P <0 .0 1) ,且对雌激素水平的影响有明显的量效关系。结论:右旋柠烯可抑制大鼠乳腺增生,降低血清雌激素水平、提高孕激素水平可能为其作用机制。
Objective: To investigate the effect of d-limonene (D-L) on the hyperplasia of mammary and the level of estrogen and progestin in serum of rat. Methods: the hyperplastic model was established by subcutaneous injection of benzhormovarine and pregnendione. Model rats were randomly divided into five groups and received different treatment. Hematoxylin and eosin (HE) staining was used to detect the histological change of mammary tissues. Radio-immunity method was used to detect the level of estrogen and progestin in serum. Results: In groups treated with DL of high, middle and lower doses, the number of mammary lobules (9.20±2.32, 6.90±1.20, 6.10±1.37), alveolus (8.20±1.75, 6.10±2.23, 5.10±1.52), and ductal epithelial layers (5.30±0.95, 4.50±1.27, 3.10±0.99) were decreased significantly compared with that of in model group (10.00±1.54, 12.91±3.88 and 6.17±1.59 respectively, P<0.05). In high dose and middle dose group, the estrogen level (28.35±2.16, 25.17±1.25) was decreased and the progestogen level (33.84±2.23, 35.39±2.20)was increased compared with model group (32.63±0.93, 31.77±1.48, P<0.05), and there exists a notable dose-effect relationship between estrogen level and the dosage of DL. Conclusions: d-limonene can inhibit the hyperplasia of rat mammary glands, and decreasing the estrogen level and increasing the progestogen level in serum may be the mechanism.
