达氟沙星对施氏鲟肝脏抗氧化功能和转氨酶活性的影响 被引量:8

摘要 分别用20、50、100mg/(kg体重)3个剂量的达氟沙星(Danofloxacin)水溶液连续口灌施氏鲟(Acipenserscherensckii)20d,并于给药后第5、10、15、20天用试剂盒测定肝脏SOD、CAT、GOT和GPT的活性及MDA和NO的含量。施氏鲟体重75~95g。结果表明,短时间内给药后(5天),100mg/(kg体重)组SOD活性与对照组相比提高显著(P<0 01)。随着实验时间的延长(第10、15天),与对照组相比各实验组SOD酶活性均保持在较高活性(P<0 01)。至第20天,各实验组SOD活性呈下降趋势,但仍明显高于对照组。给药后第5天时,各实验组CAT活性与对照组之间无差异;至第10、15天时,50、100mg/(kg体重)组均极显著高于对照组;给药后第20天,仅100mg/(kg体重)组显著低于对照组。实验第5、10、15、20天时,各实验组与对照组之间GPT和GOT活性均没有明显的差异性。仅在相同实验组内随着时间的改变有些变化。与对照组相比,在实验第5、10、15、20天各剂量组MDA含量均无显著的变化。20、50mg/(kg体重)组在第5、10、15天时有所降低,但至第20天时均恢复到对照组的水平;100mg/(kg体重)组至第15天降至最低,但实验结束时反而明显高于对照及其他实验组。从实验结果看,在实验第5、10、15、20天,各实验组施氏鲟肝脏NO含量与对照组相比较没有明显的变化。结果表明。 Danofloxacin is a kind of medicine specially used for animals.In some countries it has been used in aquaculture.The purpose of this experiment was to test the effect of danofloxacin by oral administration for 20d on the toxicity of Amur sturgeon(Acipenser scherensckii).Amur sturgeon were fed with common diets for (10d) before the beginning of this experiment.As superroxide (SOD) and catalase(CAT) are two kinds of the most important antioxidant enzymes; malondialdehyde (MDA) and nitric oxide (NO) are two kinds of the malodialdehyde; GOT and GPT are major transaminases that indicate the injured conditions of liver,the effects of danofloxacin on the activities of SOD and CAT,the contents of MDA and NO, and the activities of transominases (GOT and GPT) in crud hepatic homogenates of the Amur sturgeon were measured at different time (on 5th,10th,15th and 20th days).The Amur sturgeon were treated by the doses of 20,50 and 100mg/(kg BW) separately.The activities of SOD,CAT,GPT and GOP and the levels of NO and MDA were measured by the test kit.The values of the means of SOD,CAT,GPT,GOP,NO and MDA were analyzed by SPSS software to test significant differences.The results showed that on 5th day,the SOD activity of 100mg/(kg BW) group was higher than that in the control group significantly,and on 10th and 15th days the activities in these three test groups all remained higher value than that in the control group.At the end of experimental period the SOD activities of 20mg/(kg BW),50mg/(kg BW) and 100mg/(kg BW) groups showed lower values than those on 5th,10th and 15th days,but was still higher than that in the control group.The activities of CAT of all the three test groups were not significantly different compared to that in the control group on 5th d ;on 10th d and 15th d, the activities in 50mg/(kg BW) and 100mg/(kg BW) groups were higher evidently than that in the control group,but on 20th day only the CAT activity of 100mg/(kg BW) group was lower than that in the control group obviously.The GOT and GPT activities of 20mg/(kg BW),50mg/(kg BW) and 100mg/(kg BW) groups were not altered evidently throughout the experiment period,only in the same dose group it changed on 5th,10th,15th or 20th days.In comparison with the control group,the contents of MDA in liver homogenate of 20mg/(kg BW),50mg/(kg BW) and 100mg/(kg BW) groups remained unchanged remarkably through all the test periods.The results revealed that although on 5th,10th and 15th days the MDA contents of 20mg/(kg BW) and 50mg/(kg BW) groups were lower than that in the control group; on 20th day all remained the same level as those in the control group.The contents of MDA in 100mg/(kg BW) group was the lowest on 15th day,but on 20th day it was higher than that in the control and other test group.During the experiment period,the content of NO in liver almost was not altered in 20mg/(kg BW),50mg/(kg BW) and 100mg/(kg BW) groups.The results show that danofloxacin can affect the activities of SOD and CAT,but almost does not affect the GOT and GPT activities and the levels of MDA and NO significantly compared with the control group.The conclusion is that the effects of danofloxacin on antioxidant defenses and transaminases in Amur sturgeon liver are not obvious at the experimental dosages.Many researches showed that fluoroquinolones was a kind of medicine less toxicity for animals.Our result is also an evidence for this viewpoint.
