小鼠氧乐果亚慢性染毒对其睾丸特征性酶的影响

摘要 研究了氧乐果亚慢性染毒对雄性小鼠睾丸特征性酶的影响。将80只昆明雄性小鼠随机分成4组。染毒组分别灌胃0.5、1、2mg/kg氧乐果,对照组灌胃生理盐水,每周灌胃5d,共灌胃7周。用比色法检测睾丸组织中睾丸特征性酶——碱性磷酸酶(alkaline phosphatase,AKP)、酸性磷酸酶(acid phosphatas,ACP)、乳酸脱氢酶(lactate dehydrogenase,LDH)及乳酸脱氢酶X酶(lactate dehydrogenase–x,LDH-x)的活力,所得数据采用SPSS11.0软件进行方差分析。结果表明,随着氧乐果染毒剂量的增加,高剂量氧乐果可使部分雄性小鼠睾丸产生明显的睾丸萎缩,实质硬化等病变。睾丸AKP和ACP活力(分别为201.27±56.01U/g和348.61±78.58U/g)明显升高,与对照组(AKP为130.67±17.02U/g,ACP为239.97±40.39U/g)比较有极显著性差异(P<0.01),睾丸LDH-x(645.95±177.28U/g)的活力则显著下降(P<0.05,对照组为1058.56±159.20U/g),表明亚慢性染毒氧乐果对小鼠睾丸特征性酶有影响,并呈现明显的剂量效应关系。 This paper is designed to reveal the effect of omethoate with long-term taking on the characteristic enzyme in mice testicle. 80 male Kunming mice were divided into 4 groups: inclding one control group treated with 0.9 % NaCl solution and three groups treated with omethoate whose dose was separately 0.5,1,2 mg/kg.All mice were perfused for 5 days in every week and lasted 7 weeks. Biochemistry method was used to measure the activities of the characteristic enzymes (alkaline phosphatase,acid phosphatas, lactate dehydrogenase and lactate dehydrogenase –x,AKP, ACP, LDH and LDH-x) in mice testicle. Variance experiment data were analyzed by the software SPSS 11. The results showed that some mice,s testicles from high dose group changed with increase of omethoate dose. The testicles had obviously atrophy and the substance had ossified. The activities of the AKP and ACP in mice testicles significantly improved, which were respectively 201.27±56.01U/g and 348.61±78.58U/g, but the activities of AKP and ACP from the control group were respectively 130.67±17.02U/g,and 239.97±40.39U /g,, performing significant difference (P<0.01); and the activities of LDH-x (645.95±177.28 U/g) obviously decreased in comparison with the control group(1058.56±159.20U/g). It was concluded that the omethoate with long-term taking could effect the activities of the characteristic enzyme in mice testicle and display an obvious relationship of doseeffect.
