摘要 目的建立金银花中绿原酸的色谱分离纯化方法.方法采用亲脂性吸附树脂与葡聚糖凝胶色谱纯化技术,将其水提物经亲脂性吸附树脂柱色谱预分离后,0.1mol?L-1盐酸调节pH到4.0左右,葡聚糖凝胶sephadexLH-20精制纯化,洗脱溶剂为50%丙酮水溶液.结果产品经MS、红外、紫外和HPLC图谱鉴定,并与对照品比较对照,结果显示所得的绿原酸纯度>98%.结论该方法简单,重复性较好,可用于绿原酸单体成分的制备. Objectivel A method for the efficient preparati ve purificati on of Chlorogenic acid in the medicinal plant Lonicera japonica Thunb. has been developed. Method The separation and purification of chlorogenic aci d was achiev ed by a combination of polyamide column chromatography and sephadex LH-20 chrom atography. Water extract of Lonicera japonica Thunb. was isolated by polyamide c olumn chromatography, followed by Sephades LH-20 chromatography using a solvent system of acetone-water(5:5).Results Identification of Chlorogenic acid was performed by LC ESI MS, IR and UV-Vis. The structure of the product w as further confirmed by comparison with authentic sample and the purity of chlor ogenic acid purified was great than 98%. Conclusion This purificatio n protocol has significa ntly enhanced the throughput and simplified the isolation and purification opera tion, provided high-quality products for biological screening.
