凋亡素在体外对人卵巢癌细胞及正常卵巢细胞凋亡的影响 被引量：1

摘要 目的　探讨凋亡素基因 (VP3)表达对人卵巢癌细胞和正常卵巢细胞凋亡的影响。方法　采用LipofectamineTM2 0 0 0介导的基因转染方法将重组凋亡素真核表达质粒pcDNA3.1 -VP3 导入仓鼠正常卵巢细胞CHO -K1、人卵巢癌细胞CoC1及人卵巢癌细胞耐药亚株CoC1 DDP。结果　凋亡素基因已在转染细胞中存在并在转录水平表达 ;CHO -K1中凋亡素组与空质粒组凋亡率差异无显著性 (P >0 .0 5 )。CoC1和CoC1 DDP中 ,凋亡素组凋亡率明显高于空白对照组和空质粒组 (P <0 .0 1 ) ,且凋亡素更易诱导CoC1 DDP细胞凋亡 (P <0 .0 1 )。结论　在转染细胞中凋亡素基因在转录水平得到表达 ;凋亡素能特异性诱导人卵巢癌细胞CoC1及人卵巢癌细胞顺铂耐药亚株CoC1 DDP凋亡 ,不影响正常卵巢细胞CHO -K1 ;凋亡素更易诱导卵巢癌顺铂耐药亚株CoC1
